Metadata: Reconstructing temperature profiles for the common whelk, Buccinum undatum, using stable isotopes
Abstract:
Stable isotope data (d18O) collected from the shells of the common whelk, Buccinum undatum, from three sites (Shetland, Nefyn and Jersey) for the purpose of estimating in-situ temperatures experienced during shell calcification, from early as 2013. These estimations allowed the reconstruction of temperature profiles and calculation of growth rates over 6 month periods.

Chosen shells showed little shell damage, to prevent sampling across repaired (more recent) shell growth and maximise the number of growth cycles presented. B. undatum shells were prepared by thoroughly cleaning to remove surface debris and organic material, followed by dripping in 5% HCl to loosen remaining contaminants, followed by a scrub with a Decon90 mix with water, and then a final wash with deionised water. Shells dried in an oven at 40 °C for around 3 hours. Sequential samples were taken from the shell using a Dremel Lite 7760 Cordless Rotary Tool with a diamond point drill tip (1 mm diameter). Shallow indentations using the drill were consecutively created in the outer prismatic layer, along a single shell striation, from the lip backwards. This ensured that samples were representative of the time at shell formation. The produced powder was collected on weighing paper positioned beneath the shell. To prevent cross-contamination, the weighing paper was replaced, and the drill tip cleaned after each sample. Powder was transferred into labelled 1.5 mL Micro Centrifuge Tubes with caps and placed in an MTC™ Bio Fraction Collector Tube Rack. Approximately 100 - 250 ug of whelk material was used for isotope analysis using either an IsoPrime 100 dual inlet or an IsoPrime isoFLOW mass spectrometer at the British Geological Survey. Samples were loaded into glass vials and sealed with septa. The automated systems evacuate the vials and deliver anhydrous phosphoric acid to the samples at a set temperature. Isotope values (d13C, d18O) are reported as per mille (‰) deviations of the isotopic ratios (13C/12C, 18O/16O) calculated to the VPDB scale using a within-run laboratory standard calibrated against NBS-19. The aragonite-acid fractionation factor applied to the gas values is 1.00854. The Craig correction is also applied to account for d17O. The average analytical reproducibility of the standard calcite (KCM) is <0.1‰ for d13C and d18O. During data preparation, interpolation was carried out for samples which could not be processed due to either a sampling or analytical error. Data analyses was conducted in R (4.3.3) and a species-specific paleotemperature equation and site-specific water isotope (d18Ow) values (Hollyman et al., 2020) were used to estimate temperatures from d18Os (shell) isotope data. Dates of thermal maxima and minima, derived from hourly sea bottom temperature data for each location, were assigned to each temperature reconstruction. Dates were then interpolated for each 6 month period, allowing analyses of growth rates for individual whelk. Data analyses and project completion is still in process.
